I'd line the inside trim with 2x4's... bottom.... corners... top... frame it out on the inside

Is this stand meant to be used for aquariums? If so it should be fine just the way it is.

Should be fine as is unless there's water damage. If there is damage to it, I'd put 2x4s in the interior as support

if it were me, i would reinforce with at least a frame of 1x4s. there isnt much wood on the front and back to keep this from twisting, bear in mind a 40g tank will weigh over 350 pounds. of course, i tend to be over cautious with these types of things.

If you are concerned just put another layer of plywood in the stand....

Wood is very strong I prefer working with metal but wood will do the job for fish tank stands...

Looks fine to me. If you look at how Oceanic builds stands for their 180gal and larger tanks, you'd be surprised at how flimsy they look. But if they're good enough for Oceanic...

Just to add to the conversation, white pine wood is rated for about 4800 psi under compression if I recall correctly. Which means a square inch piece of wood could hold up my 280 gallon tank and then some if you can get it balanced on that piece just right and keep the wood from bending. Bending is the weakness for wood and thats where the extra pieces, width, and bracing comes in when building stands. But you don't need a lot of wood to hold up a tank. It's kinda funny to see ppl build stands out of 4x4 and double 2x4 etc.

That psi number is probably more towards studs than strength of plywood on end. With this stand it looks like everything is plywood. While plywood is still strong, looking at the chipping on the inside, it's not real high quality plywood either. The stand will hold the weight fine if it's built straight. Just put a level to it, if everything looks square, it will be more than fine. I'd reinforce it if it's not perfectly square though.
