I don't think the spot is something to worry about, it just looks like normal coloration.
Thanks for posting your results. Have you tested the water again? Try it and see if the nitrite values have lowered. Now that you have nitrates, the nitrites should be decreasing. If not, I would do a small water change if you notice that your fish are stressed. It may prolong the cycle, but in the long run the fish will benefit.
Are you using a dechlorinator?
