                It depends on your aquarium volume . If my memory serves me correctly, I think that Seachem suggest that you use 100ml per 100gals of water . You can go to Seachem's website to be sure .

                  I just bought a 500ml container off Amazon for like $25. And in regards to how much i used about 100-150 ml of it in my first tray with the sponge...didnt measure really. i also used the Seachem Matrix stones in the second tray of the canister.

                    Wouldn't I want it in the last tray before it goes back into the tank? Or does it matter

                        honestly i dont know if the ordering truly matters.

                        the water passes through both materials anyway. the matrix and purigen do similar functions. matrix are larger gravel size rocks that can sit directly in the basket. i laid a filter pad with carbon inside it on top of the matrix rocks too. i also have some coarse type batting separating each tray....so mine is like 6 layers from intake to out tubing (sponge, purigen, cotton material, matrix, filter pad, cotton material). hasnt impacted the flow at all

                        the purigen being much more fine in texture, i just made my own bag out of odessa that i bought at hobby lobby. also can get the cotton/batting at hobby lobby/michael's/etc

                        mine may be overkill because my Cascade 700 is under powered for a 75gal tank. until i get another filter, i am under stocking and just packing in the good media

                          On the last stage of the canister I use activated carbon and ammonia remover, it will help low ammonia and clear water put back to the tank. You can buy at Petco or Petsmart local strore ( they sell bag $1.00 so you can put carbon in one bag and another ammonia remover in one bag and it fit in one tray) In my canister I have 4 trays and I have : 1. 20 & 30ppi foam filter, 2&3 . Fluval Biomax and last one activated carbon and ammonia remover (Seachem Denitrate). It work well in my tank . If you buy online you can go to http://www.kensfish.com/ or http://www.petmountain.com/ they have good price.
